We analyzed HEK293 cells in which mtDNA was depleted by doxycycline addition to the medium to turn a dominant negative mitochondrial DNA polymerase (DN-POLG). Samples were analyzed at days 0, 3, 6 and 9 for a total of 4 samples. Each time point was done in three indepedent biological replicates. Time point 0 was used as controls with samples being un-induced. To determine the TCA cycles effects, HEK293 DN-POLG overexpressing AOX/NDI1 were also included at days 0 and 9, with 3 biological replicates to each Gene expression was also evaluated in 143B cells in which mtDNA was chronically depleted based on ethidium bromide treatment. Samples include 3 independent biological replicates of controls (rho+) and 3 of mtDNA depleted (rho0).
