Suspensions were incubated with MX at 3 concentrations with shaking at 120 rpm for 30 min at 37oC and then sampled for survival, mutagenesis, and RNA extraction. For survival, a sample was diluted 10-6, and 100 Âµl of this dilution were plated in triplicate onto VBME medium supplemented with excess biotin and histidine . For mutagenesis, 50 Âµl of the treated cells were plated in duplicate onto VBME supplemented with excess biotin and trace histidine . Plates were incubated for 3 days, and the colonies were counted by an automatic colony counter. Immediately after sampling for survival and mutagenesis, RNA was extracted and purified using the Institute of Food Research Microarray Facility protocol.
