[…] Image processing plugins for Fiji (ImageJ, NIH) and Matlab (Mathworks) were used for image registration, stitching, arithmetic operation, filtering and quantification. The calcium activity of taste cells was measured from time-lapse z-stack images acquired, typically, over the middle portion of the taste bud where cell bodies are clearly distinguished. We confirmed if the adjacent planes (2–4 μm above or below) exhibited consistent activity patterns to validate the measurement. GraphPad Prism software was used for statistical analysis.
